Les protéines de point de contrôle immunitaire

Les protéines de point de contrôle immunitaire sont des régulateurs clés de la réponse immunitaire contre les menaces tout en préservant l’auto-tolérance. Le point de contrôle immunitaire classique est constitué de récepteurs co-inhibiteurs (CTLA-4, PD-1, TIGIT, LAG-3) qui inhibent la fonction des lymphocytes T et de récepteurs co-stimulateurs (GITR, 4-1BB, OX40, CD40) qui stimulent l’activation des lymphocytes T. Les recherches sur les récepteurs co-inhibiteurs CTLA-4 (cytotoxique T-lymphocyte antigène 4) et PD-1 (protéine de mort cellulaire programmée 1) ont montré une amélioration significative des taux de survie des patients atteints de tumeurs solides métastatiques après inhibition de ces voies réceptrices. Par conséquent, les anticorps médicamenteux contre les points de contrôle immunitaires pour le cancer, les tumeurs et d'autres maladies thérapeutiques constituent un mode clé dans la thérapie du cancer.

FACS analysis shows that the binding of Human CD47 to ACHN expressing SIRP-a was inhibited by increasing concentration of neutralizing SIRP-a antibody. The concentration of Human CD47 used is 20 μg/mL. IC50=9.334 μg/mL
